Evaluation of the gastropod Radix balthica as bioindicator of cobalt exposure in outdoor mesocosms

Résumé

Most studies concerning metal interactions with gastropods focus on their soft bodies. Shells have been mainly studied with the interest to relate their metal accumulation with ambient water concentration. The present study aims to examine the kinetics of metal accumulation in gastropod soft tissue and shell, in order to better understand the role of shells in the metal handling by molluscs. To that end, the gastropod Radix balthica was exposed to increasing Co concentrations (6, 30 and 60 μg.L-1 Co) during 28 days using utdoor mesocosms (PERL, TotalEnergies, France) filled with the Gave de Pau river water in June and October 2021. Water and organisms were collected after 7, 14, 21, 28 days of exposure for analysis. For gastropod, shell measurements (length and width) were done at each sampling time. Total metal concentrations were analyzed in the whole shells and soft bodies. Finally, microchemical analyses of metals in the shells were performed using Laser Ablation Inductively Coupled Plasma Mass Spectrometry (LA-ICP-MS) and Time-of-Flight Secondary Ion Mass Spectrometry (TOF-SIMS) in order to provide data with high resolution in time and within the constitutive shell layers. At 28 days, Co accumulation in soft bodies increased 5.4-fold with the increase of Co exposure concentration from 6 to 60 μg.L-1. In those conditions, shell Co accumulation in the last growth deposit was measured using LA-ICP-MS at 0.97 ( 0.47) μg.g-1 and 1.64 ( 0.08) μg.g-1, respectively. Shell Co accumulation reached a plateau with time at each studied Co concentration. Those results will allow us to determine if gastropods can be good bioindicators of metal exposure in freshwater.
